Ann Knowles was born and raised in Southern California. She attended Georgetown University and received a BA in American Studies, with specific interest in Native American Studies. As part of her degree requirements, Dr. Knowles completed a thesis that focused on Native American healthy care delivery systems and the health care status of Native Americans and how it varies on the degree of self-determination policies that certain tribes enact. This exposure to medical care solidified an interest in pursuing medicine as a career. She attended California State University, Los Angeles, in order to get the required science courses needed to prepare for medical school.

In 1989, Dr. Knowles matriculated at the University of Nebraska Medical School in Omaha, Nebraska, and then in 1993 graduated and started a combined residency in Pediatrics and Internal Medicine. She completed the 4 year residency and continued an additional year as Chief Resident of the combined residency program. After completion of her residency, Dr. Knowles and her family moved to Washington State and began practice with Squalicum Family Medicine in 1999. Dr. Knowles is married to her husband, Jonathan, and has three children, Sam, Marle, and Jack.

Her favorite quote is from a poem her son Sam wrote for Mothers Day one year in describing his mother, ((Mom) who would like to see me in college, me finishing college and a better roof. That pretty much says it all.
